Super-Enhancement and Control of Amh Expression
نویسندگان
چکیده
In previous work it was shown that mutation of site 1 in the downstream enhancer sequence (DE) led to ablation of enhancement. Mutation of the Wilms tumour factor element (Wt), situated in the Amh promoter between the tata box and the start of translation (TSS), also led to ablation of enhancement. This suggested that these sites may be the anchor points for a specific duplex factor bridging remote DNA elements to the promoter. Mutation analysis of the DNA sequence between sections 1 and 2 of DE was carried out by site directed mutagenesis. It is reported here that site 4 lying between DE1 and DE2, plays a key role in controlling the level of enhancement.

Introduction: Epileptic neurological disorder, which is controlled with medications such as sodium valproate (one of the treatment priorities for the patients with epilepsy). Polycystic ovary syndrome (PCOS) is one of complication of sodium valproate. Ovulation in PCOs Patients is disrupted, resulting in serious complications, including endometrial hyperplasia with typical and atypical forms, i...

Background The impact of cancer therapy on the reproductive potential of patients is increasingly recognized because survival rates of patients have clearly improved in recent years. The study stressed the use of AMH may allow better prediction of chemotherapy-related risk to further fertility. MaterialsAndMethods A narrative review was performed within articles published at PubMed, Elsevier, S...
متن کاملInteraction between the Wilms tumour factor-1 element in the promoter of Amh and a downstream enhancer is required for a strong expression of the gene in pre-pubertal sertoli cells
Amh (anti-Müllerian hormone) is a single copy gene which is expressed strongly in Sertoli cells in the foetal testis and participates in the onset of sexual differentiation. Its promoter driving the expression of a reporter gene (d2EGFP) has been used to analyse the role of certain defined putative elements and a downstream enhancer element in gene expression. These experiments were carried out...
متن کاملA Model to Study the Phenotypic Changes of Insect Cell Transfection by Copepod Super Green Fluorescent Protein (cop-GFP) in Baculovirus Expression System
Background: Baculovirus expression system is one of the most attractive and powerful eukaryotic expression systems for the production of recombinant proteins. The presence of a biomarker is required to monitor transfection efficiency or protein expression levels in insect cells. Methods: The aim of this study was to construct a baculovirus expression vector encoding a copepod super green fluore...
متن کاملReduced anti-Müllerian hormone (AMH) in mares with hemorrhagic anovulatory follicles
Anti-Müllerian hormone (AMH) has been observed to decrease with the development of hemorrhagic anovulatory follicles (HAFs) in mares. Two studies were conducted to evaluate AMH concentration in mares with HAFs compared to seasonally anoestrous and cyclic mares, and to elucidate changes of AMH with the development of luteinised unruptured follicles (LUFs). In study 1, AMH and progesterone were e...
